Description

Short-term protein production within host cells.

Exogenous DNA encoding the protein is integrated into the host cell genome.

Allows regulation of protein expression by modulating the activity of the expression system through external stimuli or inducers.

The target gene is continuously expressed in mammalian cells, regardless of environmental cues or inducers.

Features

Enables rapid protein production without stable genomic integration. Methods include viral vectors, non-viral delivery, or transfection of plasmid DNA or mRNA into mammalian cells.

Protein is stably expressed over multiple cell generations. Achieved by selecting cells with successful gene integration and desired expression levels. Methods include plasmid transfection, integrative viral vectors, or site-specific integration systems.

Expression levels or timing can be controlled using chemical inducers, temperature shifts, or specific culture conditions that activate or repress the gene of interest.

Enables continuous protein production at a consistent level. Commonly used for antibody, enzyme, and hormone expression. Offers stable expression with simple operation.

When to Choose

Suitable for rapid protein production, high-throughput screening, functional assays, or when only small amounts of protein are needed. Ideal for early-stage gene expression screening or small-scale protein production.

Appropriate for long-term recombinant protein supply or production of complex proteins requiring proper folding and post-translational modifications. Used in large-scale protein production, functional studies, or drug screening.

Suitable for studies requiring controlled expression, such as evaluating the impact of expression levels on cell function, or examining dose- and time-dependent effects of drugs.

Best for large-scale production when consistent expression levels are required.

Disadvantages

Protein expression is transient and often exhibits instability over time.

Requires more time to establish and carries a risk of genomic integration affecting cell growth or phenotype.

Induction conditions and culture parameters must be optimized to achieve the desired protein expression outcome.
