Principles and Application Features of Cell Transfection

Transfection refers to a specialized technique for introducing exogenous genes into cells. With the advancement of research on gene and protein functions, transfection has become a fundamental method commonly employed in laboratory work.Transfection methods can be broadly categorized into three types: physically mediated, chemically mediated, and biologically mediated. An ideal transfection method should exhibit high transfection efficiency and low cytotoxicity. The choice of transfection technique significantly impacts experimental outcomes, and many methods require optimization of parameters such as the DNA-to-transfection reagent ratio, cell density, and incubation/detection time.

Transfection efficiency varies greatly depending on cell type.
DEAE-Dextran Method

Positively charged DEAE-dextran interacts with the negatively charged phosphate backbone of nucleic acids to form complexes that are taken up by cells.

Transient transfectionRelatively simple, reproducible results, but has some cytotoxic effects. Serum must be removed during transfection.
Virus-Mediated Method

Foreign genes are integrated into the host chromosome through infection of host cells.

Stable transfection

Suitable for hard-to-transfect cells, primary cells, and in vivo cells. However, the carried gene cannot be too large, and cells must be in the division phase.

Gene Gun MethodDNA is precipitated onto microscopic heavy metal particles, and the coated particles are then delivered into cells using a ballistic device (gene gun). The DNA is gradually released and expressed inside the cells.Transient transfectionCan be used for human epidermal cells, fibroblasts, lymphocytes, and primary cells.
