Gene editing technology refers to a series of techniques that enable precise modification and alteration of target genes. Gene editing tools include zinc finger nucleases (ZFNs), transcription activator-like effector nucleases (TALENs), and the CRISPR-Cas9 system.

The CRISPR-Cas9 system is an adaptive immune defense mechanism derived from bacteria and archaea, used to resist the invasion of foreign viruses and plasmids. It is composed of the Cas9 protein, crRNA, and tracrRNA. crRNA and tracrRNA form gRNA through local base pairing. The gRNA activates and guides the Cas9 protein to the target site (PAM). Cas9 then cleaves the target sequence near the PAM site, generating a DNA double-strand break (DSB). The cell subsequently initiates non-homologous end joining or homologous recombination repair, thereby achieving gene editing.

Homologous recombination is a targeted recombination method based on the principle of homology between DNA molecules, in which recombinases play a catalytic role to ensure precise recombination of vectors and insert fragments. For example, E. coli Red homologous recombination introduces a PCR product containing 40–60 bp of homologous sequences flanking the target gene into host cells. Utilizing the catalytic activity of the λ phage Red recombinase, the introduced linear DNA fragment undergoes homologous recombination with the specific target sequence on the chromosome (or vector), resulting in replacement of the target gene by the marker gene.
