Animal Immunization FAQs

Q1: How is the immunization effect tested?

A1: Serum samples are collected before and after immunization, and their reactivity to the immunogen is tested via ELISA. The immunogen is coated onto ELISA plates, followed by serial dilution of the antiserum containing antibodies. We can also provide antisera for the client’s in-house testing.

Q2: Are blood tests performed during the immunization process?

A2: Yes, we conduct blood tests before immunization and after the second or third booster injection. The immune response is evaluated by measuring immune serum titers using indirect ELISA.

Q3: What is the purpose of pre-immune serum?

A3: Pre-immune serum serves as an excellent negative control for various experiments.

Q4: How long does the immunization process take?

A4: Typically, the immunization process takes 2-3 months. The exact duration depends on the strength of the host's immune response.

Q5: Which species should be selected for antibody production?

A5:

SpeciesFeatures
Mouse

Advantages

1. Low cost

2. Easy to handle

3. High reproductive capacity

4. Capable of generating strong immune responses to a wide range of antigens

Disadvantages:

Limited serum yield

Rat

Advantages:

1. Multiple antibody isotypes: IgA, IgD, IgE, IgG, and IgM antibody isotypes

2. Induces immune responses to murine antigens

3. Reduced antigen requirement

4. No cross-reactivity with antigens in murine background for immunoassays

Disadvantages:

Limited serum volume

Rabbit

Advantages:

1. Capable of recognizing epitopes between rodent and human antigens that are invisible to rodent monoclonal antibodies.

2. Low cost

3. Easy to handle

Disadvantages:

Moderate serum yield

Chicken

Advantages:

1. Enhanced Immunogenicity: Due to phylogenetic distance, most mammalian proteins exhibit higher immunogenicity in chickens than in mammals, resulting in antibodies with greater affinity.

2. Superior Specificity: Chicken IgY demonstrates significantly less cross-reactivity with non-target mammalian proteins compared to mammalian IgG.

3. Low Background Interference: Structural differences in Fc regions prevent IgY from binding to IgG Fc receptors, substantially reducing false-positive staining.

4. High Yield Efficiency: Minimal antigen requirements generate sustained high-titer IgY production in egg yolks, with regular egg laying providing continuous antibody supply.

Disadvantages:

Limited antibody isotype diversity

Goat

Advantages:

1. Possesses a large amount of antiserum, 7-8 times that of small animal individual antiserum

2. Suitable for large-scale production

3. High stability

Disadvantages:

1. High cost

2. Long immunization period

Guinea pig

Advantages:

1. Higher sensitivity

2. Generates a stronger antibody response against certain antigens that are highly homologous in humans, mice, and rats.

3. Provides novel solutions for specific targets

Disadvantages:

1. High production costs

2. Prolonged immunization timeline

Camel

Advantages:

1. Novel antibody production using heavy-chain only

2. High physicochemical stability

3. Easy to construct multivalent formats

4. Rapid tissue penetration and fast clearance

5. High expression efficiency

Disadvantages:

1. Relatively long immunization period

2. Limited applicability to small molecule antigens (e.g., haptens) due to the single-domain nature of the antibodies
