scFv Antibody Library Construction Services

scFv is a small antibody fragment (~25 kDa) composed of the heavy chain (VH) and light chain (VL) connected by a short flexible peptide linker. As the most commonly used recombinant antibody type, scFv offers small size, high specificity, large-scale production capability in prokaryotic expression systems, and a complete antigen-binding site, making it widely applicable for disease diagnosis and therapy. Its small size facilitates rapid penetration into tumors for therapeutic applications. Additionally, scFv exhibits a short residence time in non-target tissues and rapid serum clearance, giving it significant potential for tumor imaging. Common scFv antibody libraries are phage display libraries; alternative technologies like ribosome display, yeast display, mammalian cell display, and bacterial display can also be used for scFv Library Construction.

KMD Bioscience possesses a mature scFv Antibody Library Construction System capable of preparing scFv libraries from immunized animal spleen cells or human B lymphocytes. Furthermore, we conduct comprehensive analysis of scFv antibody sequence information and validate it through various experiments, including EC50 Determination, Affinity Analysis, and Flow Cytometry Blocking Assays. We offer multiple phage display platforms (including M13, T4, T7, and λ phage) to meet diverse client needs and provide personalized scFv Library Construction Services. Our researchers, with years of library construction experience, excel at building different types of scFv phage display libraries, such as immune, naive, semi-synthetic, and synthetic libraries. The scFv antibody libraries we construct feature large library sizes and high diversity, enabling the generation of high-affinity scFv antibodies. We have provided scFv Library Construction Services targeting bacteria, viruses, toxins, receptors, tumor antigens, peptides, etc., for numerous domestic and international institutions. Phage Antibody Display Technology is operationally simple, has a short experimental cycle, and allows large-scale scFv expression via mammalian expression systems. This recombinant scFv antibody technology bypasses hybridoma cell preparation by directly cloning the scFv gene into the desired phagemid vector.

We specialize in connecting VH and VL with a flexible linker peptide to construct client-specified scFvs. We provide clients with various phagemid vectors, including pMECS, pComb3X, and pCANTAB 5E. We utilize strains like E. coli TG1, XL1-Blue, and ER2738, which can be expanded for phage infection. Our constructed antibody libraries achieve large sizes (up to 109 clones), exhibit good diversity and high fragment insertion rates, facilitating the screening of scFv antibodies with satisfactory affinity and specificity for clients. We also offer Expression and Purification Services for selected scFvs according to client requirements. Beyond prokaryotic expression systems, we possess various eukaryotic expression systems for antibody proteins, such as mammalian cells, yeast cells, plants, and insect cells, enabling the production of high-quality scFv antibodies. KMD Bioscience is dedicated to providing comprehensive scFv Library Construction Services to meet the diverse needs of scientists globally.
