Fab Antibody Library Construction Service

The Fab (antigen-binding fragment) is derived from full-length antibodies (e.g., IgG) through enzymatic or chemical digestion, with a molecular weight of approximately 50 kDa. As the antigen-binding domain of an antibody, Fab comprises the heavy chain variable region (VH), light chain variable region (VL), heavy chain constant region 1 (CH1), and light chain constant region (CL). Fab offers significant advantages and is widely used in disease diagnosis and therapy. Its small size enables efficient tissue penetration and rapid clearance from the bloodstream. Without the Fc fragment, Fab avoids interference in anti-Fc-mediated antibody detection. Leveraging its permeability, Fab antibodies have become indispensable tools in research applications such as Immunohistochemistry, Immunofluorescence, and Flow Cytometry.

With extensive experience in recombinant antibody research, KMD Bioscience provides professional Fab Antibody Library Construction and Preparation Services. We produce Fab fragments using two distinct methods: (i) Enzymatic digestion of whole antibodies using papain, pepsin, or ficin to generate F(ab')2 fragments, followed by reduction to yield Fab fragments. (ii) Recombinant synthesis of F(ab')2 antibody fragments, chemically reduced to obtain Fab. Most recombinant antibody fragments are currently generated via Phage Display Antibody Libraries. Fab antibody libraries offer three key advantages: (i) Rapid screening of Fab antibodies with higher efficiency than traditional monoclonal antibody methods. (ii) Easier generation of high-affinity Fab antibodies (similar to scFv antibodies). (iii) Exceptional stability during long-term storage and compatibility with standard detection antisera.

KMD Bioscience constructs diverse, large-scale Fab Antibody Libraries tailored to client needs. Our mature library systems support Fab libraries for humans and various animals, including immune, naive, semi-synthetic, and synthetic libraries. We offer multiple display platforms: (i) Phage display libraries (most requested); (ii) Ribosome/mRNA display; (iii) Yeast / Mammalian / Bacterial cell libraries. Our phage display platforms (M13, T4, T7, λ phage) enable customized Fab Library Construction. With over a decade of experience, we build Fab libraries with sizes up to 1010 and high diversity, maximizing the likelihood of high-affinity Fab antibodies. These target antigens including viruses, bacteria, toxins, receptors, tumor antigens, and peptides.

Unlike hybridoma technology (tedious, time-consuming, contamination-prone), phage antibody display simplifies operations, shortens experimental cycles, and allows large-scale Fab production via prokaryotic expression systems. We provide diverse vectors (pMECS, pComb3X, pCANTAB 5E) and host strains (TG1 E. coli, XL1-Blue, ER2738) to meet all experimental requirements. KMD Bioscience delivers comprehensive Fab Library Services to address global researchers’ needs in recombinant antibody preparation and troubleshooting.
