VHH antibodies, derived from Camelids (Camels, Llamas, Alpacas) and Sharks, consist solely of heavy chains and represent the smallest functional antibody fragments currently known, with a size of only 12-15 kDa. The camelid immune system is unique. Nn addition to conventional antibodies, their serum contains antibodies naturally lacking light chains, making them an ideal source for obtaining VHH antibodies. VHHs are characterized by their small size, excellent water solubility, stability, acid resistance, low immunogenicity, and ability to bind unique epitopes. They clear rapidly from the bloodstream and penetrate tissues effectively. These distinctive properties enable their widespread application in biosensing, affinity capture, molecular imaging, and disease diagnosis and therapy. VHHs are typically sourced from immune or naive libraries and selected using phage or yeast display technologies. Phage Display is the most common technique, enabling the production of specific VHH antibodies through appropriate screening strategies. Furthermore, VHHs are amenable to large-scale production in E. coli and exhibit unique antigen-binding characteristics compared to other recombinant antibodies.
